چکیده

Purified insecticidal crystal protein fragments (ICPFs) of Bacillus thurinqiensis encoded by crvIA(c) and cryIA(b) of this organism were shown to be lethal to key target walnut insect pests (codling moth, navel orangeworm and Indianmeal moth). One of these genes, cryIA(c), was used to transform walnut somatic embryos using a binary vector (pWB139) in which this gene was expressed as a protein fusion with the kanamycin resistancegene from bacteria. Transgenic embryos representingindividualtransformationevents were germinated to produce shoots that were maintained as micropropagated shoot clones in the laboratory. Transgenic shoots were then grafted onto seedling rootstocks and introduced into the field. DNA analysis (Southern blotting) is presented here that demonstrates that these shoots are transformed by pWB139 or by the control construct pWB149 (which does not contain the cryIA(c) gene). Insect feeding trials using in vitro grown transgenic shoots were conducted with a test insect cabbage looper, as the target walnut pests do not consume leaf materials. Statistically significant inhibition of feeding and growth was observed in one transgenic line with cabbage looper. Plants from this line have been introduced into the field and further evaluations will be performed with field grown materials.

GENETIC ENGINEERING OF INSECT TOLERANCE IN VAUruT: IMPROVED TRANSFORMATION EFFICIENCY

The genes encoding the insecticidal crystal proteins (ICPs) of BacIllus thuringiensis are good candidates for protecting walnut plants against insect pests. Ve have estimated the dose response of three important walnut pests (codling moth, navel orangeworm and Indianmeal moth) with two highly purified insecticidal proteins of Bacillus thurJ.nglensis and show that these three insects are highly ...

Genetic Engineering of Insect Tolerance in Walnut: Introduction of next Generation Constructfor High Expression of a Bt Encodedicp

Replacement trees were planted in the transgenic field plot. A total of 124 plants in the plot now includes 3 Su-2 clones with Bt (~IA(c» and Kan, 3 similar control clones without Bt, 10 Su-2 clones with Kan and GUS, 8 SF-delta clones with Kan, and 2 untransformed control clones (Su-2 and SF-delta). Seven trees (representing 3 clones) flowered in their second season in the field. Polymerase cha...

Agrobacterium-mediated Transformation of Cotton (Gossypium hirsutum) Using a Synthetic cry1Ab Gene for Enhanced Resistance Against Heliothis armigera

Cotton (Gossypium hirsutum L.)  is an important fiber crop in Iran, cultivated on 150000-200000 ha of land.  In Iran the estimated loss due to the insect pest is more than 30%. Traditionally, pests are controlled by 10-12 times spraying per growing season of environmentally harmful chemical insecticides (e.g. endosulfan and/or methosystox). In order to produce transgenic cotton resistance to in...

Improved salt tolerance in canola (Brasica napus) plants by overexpression of Arabidopsis Na+/H+ antiporter gene AtNHX1

A significant portion of the world’s cultivated land is affected by salinity that reduces crop productivity in these areas. Breeding for salt tolerance is one of the important strategies to overcome this problem. Recently, genetic engineering is becoming a promising approach to improving salt tolerance. In order to improve the yield performance of canola in saline soils, we transformed canola w...

A brief overview of field testing and commercial application of transgenic trees in China

Since the first report on transformation of Bt gene into Populus nigra was published in 1989, transgenic approaches have been widely used in breeding trees for high tolerance to insects and environmental stresses in China. Notably, field testing of the transgenic poplar with Bt was performed in 1994 and permitted to be commercialized as the first commercialized transgenic tree in the world. Up ...
